The driver of the car, a 38-year-old woman, and her back-seat passenger, a five-year-old boy, were treated by NSW Ambulance paramedics before being taken to hospital in critical conditions.
A crime scene was established by officers attached Nepean Police Area Command and forensically examined by specialist officers attached to the Metropolitan Crash Investigation Unit.
Police have now confirmed the child died overnight.
